In Vitro effect on HPRL Secretion from Decidua-Chorion,Amnion and Placental Parenchyme and Its Effects by TRH Stimulation

Abstract
In order to evaluate the main secreting site of hPRL in amniotic: fluid during pregnancy and the effects of exogenous TRH on hPRL secretion, the explants of decidua-chorion, amnion and placental parenchyma of placenta expelled from 15 normal term pregnant women were cultured in vitro. The control was one without TRH in culture medium and the experimental group was one with added synthetic TRH in different (1, 5 and 10 §­/§¢) concentrations and measured hPRL in culture medium by using radioimmunoassay.
In control group, the mean daily secretion of hPRL (mean¡¾SEM) in decidua-chorion, amnion and placental parenchyme were 107.60¡¾6.40, 10.89¡¾1.38 and 3.23¡¾0.58 ng/100 §· respectively. In experimental group with addition of 1, 5 and 10 §¶/§¢ of TRH, mean daily secretion of hPRL were 155.52¡¾6.8, 171.86¡¾8.23 and 194.82¡¾9.32 ng/100 §· in decidua, 13.72¡¾1.53, 15.86¡¾1.69 and 18.81¡¾1.91 ng/100 §· in amnion respectively. But the above patterns of secretion of hPRL was failed to be shown in placental parenchyme._ The above results showed that the main secreting site of hPRL in amniotic fluid during pregnancy is decidua-chorion but amnion is also shown to secrete a small amount of hPRL and it is also believed that TRH is involved in the control of secretion of hPRL from decidua-chorion and amnion.
